講座紹介

アトピー性皮膚炎、皮膚の角化異常症、急性および慢性腎疾患といった難治性疾患の病態を明らかにして、その診断・治療法を開発することを目標にしています。そのために、マウスや培養細胞を用いて疾患モデルを作成し、遺伝子の発現を解析し、新たな診断方法を開発すること、シーズとなる候補薬を投与することにより実臨床で使用が可能な薬を見つけることに重点を置いて研究を行っています。

研究詳細

1. 皮膚バリア機能におけるロリクリンの役割の解明

表皮は、水分の漏出や細菌・ウイルスの侵入を防ぐなど、生体にとって非常に重要なバリアとしての機能を担っています。表皮角層細胞には細胞エンベロープ（周辺帯）とよばれる細胞膜の裏打ち構造が存在しています。細胞エンベロープを構成する主成分はロリクリンとよばれ、表皮バリア機能の維持に重要な役割をはたしています。私達は、ロリクリンの機能について、ノックアウトマウスなどを用いて研究を行っています。

2. 腎障害の病態解明とアドレナリンα2受容体の役割

現在わが国において慢性腎臓病は国民病の１つに数えられており、透析導入数が増加の一途をたどっています。しかし、残念ながら慢性腎臓病に対する根本的な治療法は確立されていません。私達は、神経伝達物質であるノルアドレナリンと腎臓に発現しているアドレナリンα2受容体の役割について注目し、腎障害に対する新たな治療法を見出すことを目的に研究を行っています。

3. 皮膚におけるコネキシンの機能に関する研究

表皮角化細胞には隣り合う細胞をつなぎ、水溶性の小さいイオンや分子が通過する細胞間結合があります。これはギャップ結合とよばれています。ギャップ結合を構成する分子はコネキシンと名付けられています。コネキシン31あるいはコネキシン30.3遺伝子の変異が原因となって発症する変動性紅斑角皮症の病態機序の解析に焦点をあてることにより皮膚におけるコネキシンの機能を明らかにしたいと考えています。

4. メガリン遺伝子の発現制御に関する研究

メガリンは、腎近位尿細管、Ⅱ型肺細胞、内耳細胞など身体中の多くの部位で発現しているLow Density Lipoprotein- Related Protein (LRP) ファミリーに属する受容体の1つです。メガリンは、腎臓近位尿細管において、糸球体でろ過された低分子量タンパク質やホルモン、抗生物質などの再吸収を行っています。ファンコニー症候群などタンパク尿を呈する疾患において、メガリンタンパクの発現低下が存在します。私たちは種々の転写因子に注目して、メガリン遺伝子の発現制御を調べています。
